肾移植受者外周血辅助性T淋巴细胞17、调节性T细胞表达及其平衡关系的研究 被引量:5

摘要 目的探讨肾移植慢性排斥反应(CR)患者外周血辅助性T淋巴细胞(Thelper,Th)17、调节性T细胞(regulatoryTcells,Treg)、Th17/Treg变化及意义。方法 采用流式细胞术检测24例肾移植CR患者(CR组)和22名健康志愿者(正常对照组)外周血中的Th17占CD4+T淋巴细胞的比例(Th17/CD4+)和CD4+CD25+CD127-Treg细胞占CD4+T淋巴细胞的比例(Treg/CD4+),以及计算Th17/Treg的比值,并作比较。结果 CR组外周血中Th17/CD4+为0~3.3%(中位数0.4%),明显低于正常对照组的0~4.0%(中位数1.65%)(P<0.01)。CR组和正常对照组的外周血Treg/CD4+分别为(11.7±4.3)%、(8.0±2.6)%,CR组明显高于正常对照组(P<0.01)。CR组外周血中Th17/Treg的比值为0~0.51%(中位数0.03%),明显低于正常对照组的0~0.83%(中位数0.21)%(P<0.01)。结论 肾移植受者外周血中存在Th17/Treg失衡,Th17/Treg失衡可能参与了肾移植的免疫异常从而导致CR的发生和发展。 Objective To investigate the alteration of T helper(Th) 17 cells,regulatory T cells(Treg) and Th17/Treg in peripheral blood of renal allograft recipients with chronic rejection(CR).Methods The ratios of Th17 cells and CD4+CD25+CD127-Treg cells in CD4+T lymphocyte of peripheral blood were detected by flow cytometry in 24 renal allograft recipients with CR(CR group) and 22 healthy blood donors(normal control group).The ratio of Th17/Treg was also calculated.Results The ratio of Th17/CD4+ was 0-3.3 %(with a median of 0.4 %) in CR group,which was significantly less than 0-4.0 %(with a median of 1.65 %) in normal control group(P0.01).The ratios of Treg/CD4+T cells in peripheral blood were respectively(11.7±4.3)% in CR group and(8.0±2.6)% in normal control group.The ratio in CR group was significantly higher than that in normal control group(P0.01).The ratio of Th17/Treg was 0-0.51 %(with a median of 0.03 %) in CR group,which was significantly less than 0-0.83 %(with a median of 0.21 %) in normal control group(P0.01).Conclusion Th17/Treg imbalance exists in the peripheral blood of renal allograft recipients.It may play a potential role in the dysimmunity in renal transplantation and lead to occurrence and development of CR.
