摘要
目的观察小鼠肾缺血再灌注损伤中缺氧诱导因子-1(HIF-1)在缺血再灌注后不同时间点的的表达变化。方法采用微型动脉夹夹闭小鼠双侧肾蒂制备急性缺血/再灌注肾损伤模型,分为假手术对照组和缺血再灌注组,于缺血再灌注后4h、1d、3d分别处死。采用半定量RT-PCR法检测缺血再灌注后各时间点HIF-1 mRNA的表达变化。结果 HIF-1 mRNA表达水平在缺血再灌注后4h时开始升高并达到高峰,1d时有所回落,3d时回落到正常水平。结论肾缺血再灌注损伤后HIF-1 mRNA表达明显增高,提示缺氧诱导因子-1(HIF-1)被激活,可能与肾缺血再灌注损伤后机体对缺氧、缺血产生适应性反应有关。
Objective To explore the expression change of hypoxia inducible factor-1(HIF-1) after renal ischemia-reperfusion injury.Methods Mice were subjected to a standard renal I/R to induce acute kidney injury(AKI) after 45 min of bilateral renal artery clamping and were sacrificed at 4h,1d and 3d following injury.The mRNA expression levels of HIF-1 at different time points post injury were evaluated by RT-PCR.Results The mRNA expression of HIF-1 increased and peaked at 4h post injury,but decreased at 1d post injury,and decreased to basal level at 3d post injury.Conclusions The increased mRNA expression levels of HIF-1 post injury demonstrate that HIF-1 may be associated with the adaptive response of hypoxic or ischemic conditions after renal ischemia-reperfusion injury.
