密蒙花方对缺氧状态下脐静脉内皮细胞增殖及HIF-1α表达的影响 被引量:9

摘要 目的研究密蒙花方对缺氧状态下人脐静脉内皮细胞(human umbilical vein endothelial,HUVEC)增殖及缺氧诱导因子1α(hypoxia-inducible factor 1,HIF-1α)表达的影响。方法应用四甲基偶氮唑盐(MTT)法检测密蒙花方对缺氧状态下HUVEC增殖的影响,应用免疫细胞化学法检测密蒙花方对缺氧状态下细胞中HIF-1α表达的影响。结果在缺氧状态下,HUVEC增殖明显且细胞中HIF-1α表达增高(P<0.01),密蒙花方能有效的抑制细胞增殖及HIF-1α表达,并呈浓度依赖关系。结论密蒙花方可能通过抑制HIF-1α的表达而抑制血管内皮细胞的增殖,从而对新生血管的形成起到一定的抑制作用。 OBJECTIVE To investigate the effects of Buddleia Compound Decoction on the proliferation of HUVEC and the expression of HIF-1α in HUVEC in hypoxia state.METHODS Through MTT method the inhibition effect of Buddleia Compound Decoction on the proliferation of HUVEC in hypoxia state was checked,and through immunohistochemistry method the expression of hypoxia inducing factor-1α(HIF-1α)regulated by Buddleia Compound Decoction in HUVEC induced by chronic hypoxia in vitro was tested.RESULTS Compared with blank group,the proliferation of hypoxia group was obvious(P0.01),and the expression of HIF-1α increased in hypoxia group(P0.01).Compared with hypoxia group,the proliferation of Buddleia Compound Decoction groups was inhibited,and the expression of HIF-1α of Buddleia Compound Decoction groups was decreased.CONCLUSIONS Buddleia Compound Decoction could inhibit the proliferation of HUVEC possibly by reducing the expression of HIF-1α in HUVEC in hypoxia state.It indicates that Buddleia Compound Decoction may be useful to inhibit the neovascularization of DR.
