摘要
为探讨黄芪多糖(APS)对免疫雏鸡的免疫调节作用和机理,本研究将150羽14日龄罗曼公鸡随机均分成3组,用鸡新支二联疫苗(ND-IB)免疫的同时,2个试验组分别肌肉注射高、低剂量的APS溶液,对照组注射生理盐水,每天1次,连续3d。免疫后第10d~60d,每隔10d每组随机取10只鸡,用微量法测定血清中抗新城疫病毒血凝抑制(HI)抗体效价;每组随机取5只鸡,用流式细胞双染色法检测外周血CD3+、CD3+CD4+和CD3+CD8+淋巴细胞百分数,用ELISA方法测定空肠黏液中SIgA的含量,用免疫组织化学法测定盲肠扁桃体和十二指肠绒毛中SIgA阳性细胞数。结果显示,APS组的HI抗体效价、CD3+淋巴细胞数、CD4+/CD8+比值、SIgA的含量和SIgA阳性细胞数都明显高于对照组,且低剂量的APS的效果优于高剂量。本实验表明合适剂量APS对ND-IB疫苗的免疫效果有明显的增强作用。
This study was conducted to evaluate the effects of astragalus polysaccharide(APS) on immune responses in vaccinated chickens.A total of 150 14-day-old chickens were randomly assigned equally into three groups and all chickens were vaccinated with NDV/IBV combination vaccines.The chickens in two test groups were intramuscularly injected with APS at high(2 mg) and low dosages(1 mg) and the control group was injected with saline at the same time as the vaccination,once a day for three successive days.On days 10,20,30,40,50 and 60 post vaccination,Blood and tissue samples from in each group were randomly collected for titeration of the NDV HI antibody,determination of the proportions of CD3+,CD3+/CD4+ and CD3+/CD8+ cells,assay of the number of positive IgA secreting cells in the intestinal mucosa and cecal tonsil tissue and the secretory immunoglobulin A(SIgA) which released into the intestinal mucus.The results showed that the antibody titers,the number of CD3+ cells,the ratio of CD4+/CD8+ T lymphocytes,the concertration of SIgA and the number of the SIgA positive cells in two experiment groups were significantly higher than those in control group,and the immunoenhancement of APS at lower dosage were better than higher dosage.These findings indicated that the APS possessed a significant immune enhancing properties on NDV/IBV combination vaccines in chickens.
