摘要
目的探讨五味子多糖对环磷酰胺(Cyclophosphamide,CY)诱导小鼠胸腺细胞凋亡的保护作用及相关机制。方法小鼠腹腔注射CY,建立胸腺细胞凋亡模型,施以不同浓度五味子多糖灌胃,利用流式细胞仪、末端转移酶介导的缺口末端标记法(TUNEL染色)及Bcl-2、Bax免疫组化分别观察不同剂量的五味子多糖对CY诱导的胸腺细胞凋亡的影响及机制。结果流式细胞仪和TUNEL染色显示CY组与对照组相比的细胞凋亡率明显增高,五味子多糖治疗组与CY组比较凋亡率明显降低(P<0.01)。CY组bcl-2基因表达明显下降,bax基因表达明显增加,Bcl-2/Bax比值下降。五味子多糖组与之相反,二者比值升高(P<0.01)。结论五味子多糖能够抑制CY诱导的胸腺细胞凋亡,降低胸腺细胞的损伤,其抑制作用可能通过促进Bcl-2表达、抑制Bax蛋白表达实现的。
Objective To investigate the protective effect of polysaccharide from Fructus Schisandrae on thymocyte apoptosis induced by cyclophosphamide(CY) in mice and to explore its possible mechanism.Methods Thymocyte apoptosis model was established in mice by intraperitoneal injection of CY.After treatment with different dosages of polysaccharide from Fructus Schisandrae,thymocyte apoptosis was detected by flow cytometry,TUNEL staining,and immunocytochemistry.Results CY-induced group had a higher apoptotic rate than the normal group,and the apoptotic rate was lower in polysaccharide group than that in the CY-induced model group(P 0.01).Expression of bcl-2 gene was inhibited,while expression of bax gene was promoted,and the Bcl-2/Bax ratio was decreased in CY-induced group.Polysaccharide from Fructus Schisandrae counteracted the above changes.Conclusion Polysaccharide from Fructus Schisandrae can inhibit the apoptosis of thymocytes induced by CY and relieve the damage of thymocytes,and its inhibitory mechanism may be related with promoting expression of bcl-2 gene and inhibiting protein expression of bax.
