应用流式细胞术测定植物DNAC-值的实验方法研究 被引量:11

On method of application of flow cytometry to determine plant DNA C-value.

摘要 物种的DNA C-值具有重要的生态意义,同时也具有种的特征,目前DNA C-值越来越广泛地应用于物种倍性和杂种鉴定、植物生态适应能力分析等。本文应用流式细胞术,以油菜(Brassica campes-tris)为材料,从标样、细胞核悬液、样品生长时期等方面研究了影响DNA C-值检测的实验因素,发现标样种类和样品的生长时期对实验结果没有明显影响;与Galbraith缓冲液和Otto缓冲液相比,应用Tris-MgCl2细胞核悬液能够得到稳定的实验结果。 DNA C-value is of important ecological significance and no much variation for species.The value has been widely applied in the study in relation to species polidy and hybrid identification,and ecological adaptability,etc.The present paper brief summarized the main methods to determine plant DNA-C value,proposed a modified method to determine the DNA C-value of plant by using the method of flow cytometry,and taking Brassica campestris as experimental material.The factors influencing the determination of DNA C-value were also analyzed from internal standard,nuclei suspension,and samples growth period.We found that species and growth period of the samples have no significant effect on the experimental results,Application of Tris-MgCl2 nuclei suspension liquid could produced stable experimental results compared with other two buffers.
