摘要
以黑莓品种"Arapaho"无菌苗的叶片、叶柄为外植体,研究了植物生长调节剂、基本培养基和光照条件对愈伤组织诱导的影响。结果表明,在MS培养基中单加NAA、2,4-D、6-BA或者TDZ,都可以使黑莓"Arapaho"的叶片、叶柄产生愈伤组织,但是植物生长调节剂及其浓度不同,使出愈时间、愈伤组织诱导率以及愈伤组织大小和形态等有差异;基本培养基类型和光照时间长短对黑莓叶片、叶柄愈伤诱导无明显影响。
Experiment was carried out using leaves and petioles of blackberry 'Arapaho' as explants for evaluating the effect of plant growth regulators,dark treatments and basic media on callus induction.The results showed that calli were induced from leaf and petiole explants on MS media supplemented with NAA,2,4-D,TDZ or 6-BA.Differences were observed in induction periods,induction rates,sizes and shapes of calli,depending on hormones and their concentrations.No significant variation was found on callus induction from leaf and petiole explants of blackberry under dark treatments and different basic media.
