摘要
目的建立高效液相色谱法测定马齿苋中α-亚麻酸、亚油酸的含量。方法色谱柱:Shim-pack CLC-ODS(4.6 mm×250 mm,5μm);流动相:甲醇-乙腈-0.5%磷酸水溶液(66∶22∶12);柱温:26℃;检测波长:210 nm;流速:1.1 ml/min。结果α-亚麻酸的回归方程为A=2.394×107C-2.623×103(r=0.9999),线性范围:1.60~64.00μg/ml;亚油酸的回归方程为A=1.066×106C-2.927×103(r=0.9998),线性范围:1.68~67.20μg/ml。结论不同产地、批次的马齿苋中α-亚麻酸、亚油酸的含量差异较大。现行版中国药典尚无马齿苋药材的含量测定项目,有必要将亚麻酸、亚油酸含量指标纳入马齿苋药材的质量评价体系。
Objective To develop an high performance liquid chromatography method for the determination of α-linolenic acid and linoleic acid in Portulaca oleracea L.Methods The method was developed under the following conditions: Shim-pack CLC-ODS column(4.6 mm×250 mm,5 μm) was used with the mobile phase of methanol-acetonitrile-0.5% phosphonic acid(66∶22∶12)at a flow rate of 1.1 ml/min.Column temperature was at 26℃ and the detection wavelength was set at 210 nm.Results The standard curves of α-linolenic acid and linoleic acid were linear in the range of 1.60-64.00 μg/ml and 1.68-67.20 μg/ml,respectively.The standard curves of α-linolenic acid and linoleic acid were A=2.394×107C-2.623×103(r=0.9999) and A=1.066×106C-2.927×103(r=0.9998),respectively.Conclusion The content of α-linolenic acid and linoleic acid in Portulaca oleracea L.growing in different districts and of different batches.For no quantitative analysis item of Portulaca oleracea L.included in China pharmacopoeia(2005 year edition),it is necessary to include α-linolenic acid and linoleic acid in quality evaluation system of Portulaca oleracea L.
