摘要
目的:采用共振瑞利散射法(Resonance Rayleigh scattering,RRS)测定不同制剂中阿米卡星(Amikacin,AMK)的含量。方法:考察RRS增强的机理、反应条件对RRS强度的影响以及AMK浓度与RRS强度的关系。结果:在pH3.0的Britton-Robinson缓冲液中,滂胺天蓝(Pontamine sky blue,PSB)与AMK在静电引力及疏水性作用结合成组成比为2∶1的离子缔合物。AMK的浓度在0.01~1.7 mg/L范围内与RRS强度成正比,检出限(3σ)为3.0μg/L。结论:该法简便快速、结果准确可靠,可用于不同制剂中AMK的含量测定。
Objective:To determine simply and rapidly amikacin(AMK) in different pharmaceutical formulations with resonance Rayleigh scattering(RRS) method.Methods:The mechanism of generating greatly enhanced RRS signals,the effects of the interacting conditions on the enhanced intensity of RRS,and the relationship between the enhanced RRS intensity and the concentration of AMK were investigated.Results:In a Britton-Robinson buffer solution(pH3.0),AMK interacted with pontamine sky blue(PSB) to form an ion-association complex with the composition ratio of 2∶1 by virtue of electrostatic attraction and hydrophobic effect.It was found that the enhanced intensity of RRS was in good proportion to the concentration of AMK in the range of 0.01~1.7 mg/L with the detection limit(3σ) at 3.0 μg/L.Conclusion:The method is satisfactorily applied to the determination of AMK in different pharmaceutical formulations.
