摘要 目的通过体外培养人脂肪细胞,研究C-反应蛋白对脂肪细胞脂联素蛋白分泌及mRNA基因表达的影响。方法取女性皮下脂肪组织培养脂肪细胞。用01、05、0μg/mL的C-反应蛋白刺激脂肪细胞6 h,提取细胞RNA用实时荧光定量RT-PCR技术检测脂联素mRNA表达的变化;收集细胞培养液,运用Western-blot技术检测脂联素蛋白分泌的变化。结果 50μg/mL的C-反应蛋白可提高人脂肪细胞脂联素mRNA的表达(P<0.05),10μg/mL的C-反应蛋白对人脂肪细胞脂联素mRNA的表达无影响(P>0.05)。50μg/mL、10μg/mL的C-反应蛋白均可减少人脂肪细胞培养液中脂联素蛋白的分泌量(P<0.05),C-反应蛋白可呈剂量依赖性的降低脂肪细胞脂联素蛋白分泌的水平。结论 C-反应蛋白对脂肪细胞脂联素基因表达和蛋白分泌的研究可以揭示转录和转录后的控制决定了C-反应蛋白对脂联素的影响。 Objective To investigate the effects of C-reactive protein(CRP) on mRNA production and protein secretion in human adipocytes in vitro.Methods Obtained the adipose tissue and cultured human adipocytes.Adipocytes were incubated with concentration of CRP ranging from 0 to 50 μg/mL for 6 hours.Then the medium of the adipocytes was collected and the secretion of adiponectin in the medium was determined by using Western blot.Adiponectin mRNA was measured by quantitative real-time reverse transcription-polymerase chain reaction.Results 50 μg/mL CRP up-regulated the mRNA expression of adiponectin in human adipocytes(P〈0.05).Compared with control group,the secretion of adiponectin was inhibited by CRP(10 μg/mL or 50 μg/mL) in human adipocytes(P〈0.05).Adiponectin secretion was inhibited by different concentrations of CRP in a dose-dependent manner.Conclusion Investigating the effect of CRP on adiponectin mRNA production and protein secretion suggests that transcriptional and posttranscriptional mechanisms are determinants of regulation of CRP on adiponectin.
