摘要
目的:观察丹蛭降糖胶囊对小剂量链脲佐菌素(STZ)加高热量饮食诱导的2型糖尿病大鼠肾皮质ET-1mRNA表达的影响。方法:雄性Wistar大鼠48只,随机分为4组:空白组(B组)、模型组(M组)、马来酸罗格列酮组(R组)、丹蛭降糖胶囊组(T组),每组12只。空白组喂以普通饲料,其余3组尾静脉注射小剂量STZ并喂以高热量饲料,常规测定各组治疗前后空腹血糖(FPG)、餐后2h血糖(2hPG),空腹血清胰岛素(FIns)并计算胰岛素敏感指数(ISI),提取肾皮质总RNA,采用逆转录PCR技术扩增ET-1基因片段,检测其表达水平。结果:丹蛭降糖胶囊能够降低模型大鼠的FPG和2hPG,降低FIns,提高ISI,并能够降低ET-1mRNA的表达。结论:丹蛭降糖胶囊可减少ET-1mRNA的表达,提示其机制可能与辅助降糖、增加胰岛素敏感性、改善胰岛素抵抗有关。
Objective:To explore the effects of Danzhijiangtang capsule(DJC)on the mRNA expression of ET-1 in rats with type-2 diabetes mellitus.Methods:48 male Wistar rats were randomly divided into four groups:blank control group(Group B)、model control group(Group M)、rosiglitazone maleate treatment group(Group R) and DJC treatment group(Group T),each group with 12 rats.The blank control group was fed with ordinary diet,the other groups were fed with high-calorie diet.After 4 weeks,inducing rat model of T2 DM by intraperitoneal injected small-dose streptozotocin.To observe FPG、2hPG、Fins、ISI.To extract total RNA of kidney cortex,then to amplify ET-1 gene fragments by RT-PCR.Results:DJC can decrease FPG、2hPG、FIns,can increase ISI,also can decrease the mRNA expression of ET-1.Conclusion:DJC can significantly defend impaired vascular endothelial and improve PTS,the mechanism probably are:reducing blood sugar、improving insulin resistance and reducing the mRNA expression of ET-1.
