摘要
目的研究克糖特对链脲霉素致糖尿病(DM)小鼠的降血糖作用并初步探讨其机制。方法小鼠腹腔注射链脲霉素(STZ)150mg/kg建立DM模型,将建模成功的小鼠随机分为5组。格列本脲组:格列本脲50/(kg·d)灌胃(i.g.),克糖特高、中、低剂量[i.g.克糖特分别相当于生药50g/(kg·d),25g/(kg·d),12.5 g/(kg·d)];DM模型组:等容积生理盐水i.g.;与空白对照组:等容积生理盐水i.g.;均连续给药15d。并在相应时间采血测定空腹血糖(FBG)、胰岛素、三酰甘油(TG)、总胆固醇(TC)、血清肌酐(Cr)、尿酸(UA)等指标。结果克糖特有明显的降血糖作用,其作用呈剂量依赖性,并提高胰岛素水平;同时能不同程度地降低血清Cr、UA(P<0.01);血脂中的(TG)水平有降低趋势。结论克糖特能降低链脲霉素所致DM小鼠的血糖,增加胰岛素的分泌,对DM尤其并发症的治疗有潜在的应用价值。其降血糖作用机制可能与促进胰岛素分泌有关。
Objective To investigate the hypoglycemic effects and its mechanism of Ketangte(KTT) in streptozotocin(STZ)-induced diabetic mice.Methods Diabetic model mice were prepared by injected 150mg/kg STZ.Then the mice were randomly divided into five groups include model group,glibenclamide group[50mg/(kg·d),intragastric administration(i.g.)],KTT high dose group [50g/(kg·d),i.g.],KTT middle dose group[25g/(kg·d),i.g.] and KTT low dose group [12.5g/(kg·d),i.g.].While the control and model group were administrated with the same volume of saline for 15 days.During experiment,the FBG,insulin,TG,TC,Cr,UA were measured in different times.Results KTT could reduce the blood glucose concentration significantly in STZ-induced diabetic mice with dose-dependent effect.KTT could raise insulin levels,and decrease Cr and UA levels in serum(P0.01).Triglyceride(TG) in blood lipid had a lowering tendency.Conclusion The results suggest that KTT can decrease blood glucose,improve insulin levels in STZ-induced diabetic mice,indicating that KTT maybe have a potential value for the treatment of DM and DM complications.The mechanism of reducing blood glucose is probably associated with promoting insulin secretion.
