摘要
目的建立HPLC测定奈米非肽中三氟醋酸含量的方法。方法色谱柱:Apollo C18(4.6mm×250mm,5μm);流动相:流动相A 0.7%磷酸水溶液(PH3.0),流动相B甲醇;流速:1.2ml/min;检测波长:210nm;进样量:10μl。结果在12.0-961.2μg/ml浓度范围内,呈良好线性关系,线性方程为A=0.232 2X-0.062 6,r=0.999 6,平均回收率100.7%,RSD为0.73%(n=6)。结论本法简单可靠,重现性好,可作为奈米非肽中三氟醋酸含量测定方法。
Objective To establish the HPLC method for measurement of trifluoroactic acid level in nemifitide.Methods Chromatogram with an Apollo C18 column(4.6mm×250mm,5μm),mobile phase with a 0.7% phosphoric acid aqueous solution(pH3.0),methanol with a flow velocity of 1.2ml/min,test wavelength at 210nm,and an injection dose of 10μl,were used to establish the HPLC method for measurement of trifluoroactic acid level in nemifitide.Results The method had a good linear relation when the regression equation was A=0.232 2X-0.062 6 and r=0.999 6 at the concentration of 12.02-961.2μg/ml.The average recovery rate was 100.7% and the RSD was 0.73%(n=6).Conclusion The method is simple and reliable with a good repeatability,and can thus be used in measurement of trifluoroactic acid level in nemifitide.
