摘要
采用线栓法制作大鼠大脑中动脉闭塞(MCAO)所致脑缺血模型,研究甲磺酸胺银杏内酯B(XQ-1H)灌胃给药(ig)对局灶性脑缺血的治疗时间窗。将3批动物造模后分别于缺血后1,2,3 h给药,24 h后分别进行大鼠局部脑血流、神经行为学评分、脑梗死率、脑含水量测定以及组织形态学观察。结果显示:在脑缺血后1 h、2 h进行XQ-1H治疗给药能使局灶性脑缺血大鼠局部脑血流量增加、神经行为明显改善,脑梗死率及脑含水量显著下降,XQ-1H于脑缺血后1 h、2 h给药对局灶性脑缺血具有显著的治疗作用。表明其灌胃给药对局灶性脑缺血的治疗时间窗为脑缺血后2 h。
Dimethylaminoethyl ginkgolide B mesylate(XQ-1H)therapeutic time window in rat focal cerebral ischemia was investigated using rat model of middle cerebral artery occlusion(MCAO,induced by intraluminal suture method).Following MCAO,different doses of XQ-1H were administered(ig)at 1,2 and 3 h,respectively.Regional cerebral blood flow,neurological score,infarction rate,water content and pathological change of rats at each interval were determined 24 h after MCAO.It was observed that the regional cerebral blood flow was increased and neurological score,cerebral infarction rate and water content were reduced significantly at 1 h,2 h(XQ-1H administered after MCAO).The results showed that the neuroprotective effect of XQ-1H on the therapeutic window in focal cerebral ischemia injury of rats was extended to 2 h.
