摘要
背景:寻找一种同种异体组织保存液,使保存的肌腱具有最大程度的生物活性。目的:使用无水甘油在常温下对兔子肌腱进行保存,观察不同时间点的细胞形态和肌腱活性。方法:使用特殊的方法处理兔肌腱,并常温避光密封保存在无水甘油中,在保存2,4,7,12个月使用苏木精-伊红染色石蜡切片、透射电镜观察肌腱组织结构及细胞形态,使用超氧化物歧化酶酶学实验观察肌腱的生物学活性。结果与结论:2,4,7,12个月苏木精-伊红染色石蜡切片示大部分细胞细胞膜完整,细胞核规则,肌腱组织结构存在。透射电镜结果示细胞呈静止状态,核形态正常。超氧化物歧化酶酶学实验示肌腱具有超氧化物歧化酶酶活性。结果说明使用特殊的方法处理兔肌腱并在无水甘油中常温避光保存12个月,肌腱的组织结构、大部分细胞的细胞形态和肌腱生物活性得以保存。
BACKGROUND:In order to preserve the greatest bioactivity of the tendon,a kind of allograft preserving liquid is needed.OBJECTIVE:To observe the cell morphology and tendon bioactivity at different time points,followed by protection of the rabbit tendon in pure glycerine under normal temperature.METHODS:After special procession,the rabbit tendons were tightly sealed in pure glycerine away from light under normal temperature.The organizational structure and cell morphology of tendon were observed under electron microscope with hemotoxylin and eosin(HE) staining for paraffin section,and the bioactivity was observed by superoxide dismutase zymetology experiment for 2,4,7 and 12 months,respectively.RESULTS AND CONCLUSION:HE stained paraffin section showed that in 2,4,7 and 12-month glycerine group,most cell membranes were intact,nucleuses inerratic,and tendon organizational structures existed.The electron microscope showed the cells were in static,and the nucleus patterns were normal.The superoxide dismutase zymetology experiment showed that the tendon owned the superoxide dismutase enzymatic activity.The results indicated that the organizational structure,most of cell morphology and bioactivity of the tendon were remained,following the tendon was specially processed and tightly sealed in pure glycerine away from light under normal temperature for 12 months..
