摘要
目的观察实验性肝纤维化过程中间质胶原酶(MMP-13)及其组织金属蛋白酶抑制因子-1(TIMP-1)基因表达的不平衡性。方法建立CCl4中毒性大鼠肝纤维化模型,采用SABC免疫组化方法和逆转录定量PCR方法测定MMP-13和TIMP-1的表达情况。结果MMP-13和TIMP-1在正常大鼠肝组织中有表达,在肝纤维化发生发展过程中MMP-13表达无显著性变化,而TIMP-1的表达则逐渐增强,在肝硬化阶段达到最高值。结论MMP-13和TIMP-1在肝纤维化过程中的不平衡表达可能是肝硬化形成的重要决定因素。
Objective To investigate the imbalance in expression betWeen interstitial collagenase (MMP-13 in rat) and inhibitor. itsmetalloproteinase-1 (TIMP-1 ) gene in the processes of experimental liver fibrosis. Methods CCI. induced animal models of liverfibrosis was established. MMP-13 and TIM'P-I gene expression levels in the difl'erent stages of liver fibrosis were measured b}'imrnunohistochemistry (SABC assay) and quantitative analysis of reverse transcription polymerase chain reaction (RT-PCR).Results MMP- 13 and TIMP- 1 gene expressions could be detected in normal rat liver .In the pathogenesis and development of liverfibrosis. MMP-13 gene expression level was increased gradually. and reach peak levels at fibrosis stage. Conclusion MM-P-13 andTIMP- 1 might play an important role in the processes of pathogenesis, development of experimental liver fibrosis.
