摘要
为了筛选棉花中对绿盲蝽为害产生防御反应的基因,初步构建了绿盲蝽刺吸胁迫诱导棉花抑制性消减杂交正向文库,构建文库过程中随机挑取960个克隆进行菌落PCR鉴定,插入片段在250~1500 bp之间。经反向Northern筛选,测序拼接后得到78个非重复独立基因(unigenes)。通过Blast2GO软件对unigenes进行序列比对,并对差异基因进行功能分类,结果表明,由绿盲蝽刺吸胁迫诱导的棉花应激反应过程包括细胞通讯、酶活性调节、对外界生物和非生物胁迫的应激反应、次级代谢、能量合成代谢以及细胞生长发育等很多生物过程。这些生物过程的变化直接或间接对棉花本身起到一定的保护作用。
In order to select cotton defense gene against Apolygus lucorum Meyer-Dür(green plant bugs),we constructed the cotton forward suppression subtractive hybridization library by the green plant bugs sucking induced.960 colonies were randomly picked to colony PCR in the library,and the fragment was inserted between the 250-1500 bp.After reverse Northern hybridization and sequenced,we obtained 78 independent genes(unigenes).Through similarity research by Blast2GO software,the results showed that stress process by the green plant bugs sucking,including cell communication,enzyme activity regulation,biotic and abiotic stress response,secondary metabolism,energy metabolism,cell growth and development and many other biological processes.These processes can directly or indirectly play an important role to protect cotton against green plant bugs.This study was designed to screen defense gene against green plant bugs of cotton,and provided a new way and method to prevent and control green plant bugs.
