摘要
目的研究苦参碱对急性心肌缺血大鼠心肌梗死范围和心肌超微结构的影响。方法将SD大鼠随机分为7组:正常对照组、假手术组、模型组、硝酸甘油组,苦参碱2.5、5、10mg/kg剂量组。应用左冠状动脉前降支结扎法制备大鼠急性心肌缺血模型,TTC染色法测定心肌梗死面积,电镜观察心肌细胞超微结构的改变。结果静脉注射苦参碱2.5、5、10mg/kg,可减小急性心肌缺血大鼠心肌梗死区重量和梗死百分率(P<0.05或P<0.01),改善心肌细胞线粒体、肌原纤维、闰盘等细胞器的损伤。结论静脉注射苦参碱可通过缩小心肌梗死范围、改善心肌细胞超微结构,参与急性缺血心肌的保护作用。
Objective To investigate the effects of matrine on infarct size and ultra microstructure in rats with acute myocardial ischemic(AMI) injury.Methods Sprague-Dawley rats were divided into seven groups: control,sham operation,AMI,nitroglycerin+AMI,AMI+MT2.5mg/kg,5mg/kg,or 10mg/kg group.Rat models of AMI were established by ligation left anterior descending coronary artery.Infarct size was checked with triphenyl tetrazolium chloride(TTC) staining.Heart's ultra microstructure was observed by electric microscope.Results The infarct size of left ventricle and the ratio of the weight of infarct myocardial tissue to the whole left ventricle were significantly reduced in AMI rats after MT injection at the dosage of 2.5,5,10 mg/kg(P0.05 or P0.01).The injury of organelles(mitochondria,myofibrilla,intercalary disc,etc) was improved significantly in AMI rats after MT administration at the dosage of 2.5,5,and 10 mg/kg.Conclusion Intravenous matrine can reduce infarct size and improve ultra microstructure to protect the ischemic myocardium in rats.
