摘要
目的研究汉黄芩素对TRAIL诱导的A549细胞增殖抑制及凋亡的影响。方法将不同浓度的汉黄芩素和/或TRAIL作用于人肺癌细胞系A549细胞,MTT法检测细胞增殖率,流式细胞术Annexin V-FITC检测细胞凋亡。结果 IC50浓度的汉黄芩素联合TRAIL(100 ng/mL)及单药TRAIL分别作用于A549细胞24 h,联合组细胞增殖率明显低于TRAIL单药组(34.56%±0.52%vs 94.32%±3.28%,P<0.01);联合组细胞凋亡率明显高于TRAIL单药组(28.72%±3.66%vs 7.62%±1.05%,P<0.01)。结论汉黄芩素能够增强A549细胞对TRAIL诱导凋亡的敏感性。
Objective To investigate the effect of wogonin on TRAIL-induced apoptosis in A549 cells. Methods A549 cells were cultured and treated with wogonin and/or TRAIL.Cell viability was determined by MTT assay.Apoptosis was detected by flow cytometry. Results The A549 cells were treated with IC50 doses of wogonin and/or 100 ng/mL TRAIL for 24 h,compared with cells treated with TRAIL alone,cell viability was obviously lower and apoptosis rate was obviously higher in cells cotreated with TRAIL and IC50 doses of wogonin(cell viability:34.56%±0.52% vs 94.32%±3.28%,P0.01;apoptosis rate:28.72%±3.66% vs 7.62%±1.05%,P0.01). Conclusion Wogonin can enhance TRAIL-induced apoptosis in A549 cells.
