摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)中3个干扰素(IFN)诱导基因(IFITM1,IF-ITM2和IFITM3)的表达及其与SLE疾病活动度的相关性。方法运用逆转录聚合酶链反应(RT-PCR)方法检测30例SLE患者与25名健康对照者的IFITM1、IFITM2、IFITM3mRNA表达水平,并且分析它们与补体C3、C4、抗dsDNA抗体及SLE疾病活动指数(SLEDAI)积分的相关性。结果 (1)SLE组与正常对照组相比,IFITM1、IFITM2、IFITM3mRNA表达显著增高(分别为P<0.0001,P=0.003,P=0.002);(2)IFITM1、IFITM2、IFITM3mRNA表达水平之间呈显著正相关;(3)SLE组IFITMl、IF-ITM3mRNA表达水平均与补体C3、C4呈显著负相关(r<-0.5,P<0.05),与dsDNA、SLEDAI积分不相关。IFITM2与补体C3、C4、dsDNA及SLEDAI积分均无相关性。结论 SLE患者IFITM1、IFITM2、IFITM3的表达水平在SLE患者中显著增高,并且IFITMl、IFITM3与补体C3、C4呈显著负相关,对SLE患者的诊断和病情评估可能有一定价值。
Objective To investigate the expression levels of interferon-inducible genes(IFITM1,IFITM2,IFITM3)in the peripheral blood mononuclear cells (PBMCs) of patients with systemic lupus erythematosus(SLE),and the relations between these genes expression levels and disease activity are explored.Methods Reverse transcription polymerase chain reaction(RT-PCR)method was used to detect the expression levels of IFITM1,IFITM2,IFITM3 in 30 patients with SLE and 25 controls.The associations between the expression levels of IFITM1,IFITM2,IFITM3,complement C3,C4,anti-double stranded DNA antibody and SLEDAI scores in patients with SLE were analyzed.Results (1)The expression levels of IFITM1,IFITM2,IFITM3 in the SLE patients were significantly higher than those of the normal controls(P〈0.0001,P=0.003,P=0.002 respectively).(2)The expression levels of IFITM1,IFITM2 and IFITM3 showed positive correlations with each other(P〈0.05).(3)The expression levels of IFITM1 and IFITM3 were negatively correlated with complement C3,C4(r〈-0.5,P〈0.05),in the same time,they were not correlated with SLEDAI scores and anti-double stranded DNA antibody (r〈0.5,P〉0.05).There was no correlation between complement C3,C4,SLEDAI scores,anti-double stranded DNA antibody and the expression levels of IFITM2.(r〈0.5,P〉0.05).Conclusions The expression levels of IFITM1,IFITM2 and IFITM3 in patients with SLE are significantly higher than those of the normal controls.The expression levels of IFITM1,IFITM2 and IFITM3 showed positive correlations with each other.IFITM1 and IFITM3 were negatively associated with complement C3,C4,and they are helpful in evaluating SLE disease activity and severity.
