摘要
目的:用MTT法检测重组人角质细胞生长因子-2(rhKGF-2)诱导不同细胞系增殖的差异,建立验证rhKGF-2生物活性的细胞模型。方法:利用载体pBV220-KGF-2原核表达和纯化rhKGF-2,然后用MTT法分别检测rhKGF-2诱导大鼠气管上皮细胞RTE、大鼠小肠上皮细胞IEC-6、小鼠成纤维细胞NIH/3T3和人喉癌细胞Hep-2增殖的能力。结果:rhKGF-2具有强促Hep-2细胞增殖的生物活性,而对RTE细胞、IEC-6细胞、NIH/3T3细胞的诱导增殖作用不明显。结论:在体外,rhKGF-2具有较强的诱导Hep-2细胞增殖的作用。本研究为确立验证rhKGF-2生物活性的细胞模型的选择提供了实验依据。
Objective:To detect the proliferation disparity of different cell lines by recombinant human keratinocyte growth factor-2(rhKGF-2) stimulation with MTT experiment and to establish the cell sample to identify rhKGF-2 bioactivity.Methods:After expression and purification of rhKGF-2 in prokaryotic system,RTE cell,IEC-6 cell,NIH/3T3 cell and Hep-2 cell were prepared to verify the ability of rhKGF-2 to stimulate proliferation of cell lines with MTT experiment.Results:rhKGF-2 had a strong biological activity of promoting Hep-2 cell to proliferate,but to RTE cell,IEC-6 cell and NIH/3T3 cell,its funcation of inducing proliferation was not obvious.Conclusion:rhKGF-2 can induce proliferation of Hep-2 cells in vitro,this study provided a cell model for the establishment of verification rhKGF-2 biological activity.
