摘要
以富含γ-氨基丁酸(GABA)的大豆胚芽乳、黄酒、有机糙米、绿茶和杨梅为研究对象,采用2,4-二硝基氟苯(FDNB)柱前衍生和等度洗脱,建立一种快速测定γ-氨基丁酸含量的RT-HPLC方法。该方法简便,分析时间短(〈7 m in),灵敏度高,稳定性和重现性好。优化后色谱条件如下:HypersilODS2 C18,流动相为乙腈∶水∶磷酸盐缓冲液(pH=7.2,体积比为20∶20∶60),检测波长为360 nm。在最适条件下,γ-氨基丁酸的线性检测范围为0.016~1.000 mg/mL,线性关系良好,回收率为96.5%~104.0%。
In this paper,a method for determining γ-aminobutyric acid in Chinese bayberry was developed.It was based on a precolumn derivatization of GABA with 2,4-dinitrofluorobenzene(FDNB) and isocratic elution by RT-HPLC.It was simple,rapid(less than 7 minutes),steady,sensitive and accurate.The optimized chromatographic conditions were as follows:Hypersil ODS2 C18,eluent consisted of 20% acetonitrile,20% water and 60% phosphate buffer solution.GABA was determined with UV detector at 360nm,and good linearities were observed within a range from 0.016 to 1.0 mg/mL.The recoveries were 96.5%~104.0% for GABA.
