摘要
[目的]通过研究阿托伐他汀对LPS诱导的人血管内皮细胞TLR4 mRNA和蛋白质表达的影响,探讨其抗炎机制。[方法]采用1640培养液培养人血管内皮细胞;TLR4诱导的人血管内皮细胞分为五组:空白组(A组)、LPS组(B组)、阿托伐他汀钙低浓度组(C组)、阿托伐他汀钙中浓度组(D组)和阿托伐他汀钙高浓度组(E组),每组9个副孔,采用RT-PCR方法观察各组TLR4mRNA表达情况;用Westen Blot方法检测TLR4蛋白表达情况。[结果]与空白组(A组)相比,LPS组(B组)TLR4mRNA和蛋白质表达显著增加;阿托伐他汀抑制LPS诱导的TLR4 mRNA和蛋白质表达均呈剂量依赖性;阿托伐他汀高浓度组(E组)LPS诱导TLR4 mRNA和蛋白质表达明显减少(P<0.05)。[结论]阿托伐他汀能够通过抑制TLR4信号转导通路,抑制TLR4表达起到抗炎作用,进而发挥延缓动脉粥样硬化进程作用。
[Objective] To explore the anti-inflammatory mechanism through the study on the influence of atorvastatin on expression of LPS-induced human vascular endothelial cells TLR4 mRNA and protein.[Methods] culture human vascular endothelial cells by culture medium 1640.Human vascular endothelial cells induced by LPS are divided into five groups:blank group(A group),LPS group(B group),low dose group of atorvastatin(C group),middle dose group(D group) and high dose group(E group),each group has nine accessory foraminas,the expression of TLR4mRNA in each group is observed by RT-PCR;the expression of TLR4 protein is detected by Westen Blot.[Results] Compared with the blank group,the expression of TLR4mRNA and TLR4 protein in LPS group increases significantly;atorvastatin inhibited LPS-induced expression of TLR4 mRNA and protein exists in a dose-dependent manner;High concentrations of atorvastatin can reduce the expression of TLR4 mRNA and protein induced by LPS(P0.05).[Conclusions] Atorvastatin can make an anti-inflammatory effect by inhibiting the signal transduction pathway and expression of TLR4 and thus play a role in delaying the process of atherosclerosis.
