宁心痛颗粒干预模型兔动脉粥样硬化易损斑块机制研究 被引量:13

Mechanism study of Ningxintong Granule on interposing effect on atherosclerosis vulnerable plaques in rabbits

摘要 目的:观察宁心痛颗粒对动脉粥样(AS)易损斑块的稳定作用,探讨宁心痛颗粒稳定易损斑块的机制。方法:采用球囊损伤腹主动脉加高脂饲料喂养结合转染p53基因、中国斑点蝰蛇毒和组胺触发建立兔AS易损斑块模型,分别予宁心痛颗粒、血脂康胶囊干预,以普通病理及免疫组化观察斑块形态及NF-κB、MCP-1、MMP1的局部表达;采用Western blot技术检测NF-κB、MCP-1、MMP1水平,进行对比分析。结果:宁心痛颗粒与血脂康胶囊均能够增加纤维帽厚度及纤维帽厚度/IMT比值,降低IMT(P<0.05),两药物作用相当。免疫组化染色结果显示易损斑块模型组AS斑块处均有NF-κB、MCP-1、MMP1的局部表达,表达强度较空白组明显增强(P<0.05),宁心痛颗粒高剂量与血脂康胶囊均能够降低NF-κB、MCP-1、MMP1的表达水平(P<0.05),宁心痛颗粒高剂量与血脂康胶囊作用相当。Western blot检测模型组斑块局部NF-κB、MCP-1、MMP1蛋白水平明显增高(P<0.05),宁心痛颗粒与血脂康胶囊均能够降低NF-κB、MCP-1、MMP1蛋白水平,血脂康胶囊组NF-κB、MMP1水平略低于宁心痛颗粒高剂量组,宁心痛颗粒高剂量组NF-κB、MMP1水平低于宁心痛颗粒常规剂量组(P<0.05)。结论:宁心痛颗粒能够通过抑制炎症反应,从而达到稳定AS易损斑块的目的。 Objective:To investigate the effect of Ningxintong Granule(NXTG)on stabilizing vulnerable plaques and to explore the mechanisms of drug treatment with NXTG in stabilizing vulnerable plaques.Methods:42 healthy male new Zealand white rabbits were randomized into the group A and the group B,rabbits in group B were divided into four subgroups randomly(B1,B2,B3,B4).Rabbits in group A were fed standard chow.Rabbits in group B were fed high cholesterol diet for 2 weeks and then underwent ballon-induced abdominal aortic wall injure.At the 3rd week,group B1 was NXTG low dose treatment group,B2 was NXTG high dose treatment group,group B3 was administered Xuezhikang,group B4 was baseline(n=9).Rabbits in group B were still fed the cholesterol-rich diet.At the end of the 8th week,recombinant adenovirus carrying p53 genes were injected through a catheter into the aortic segments rich in plaques in group B.Two weeks later,the remaining rabbits in group B all underwent pharmacological triggering with injection of Chinese Russell's viper venom(CRVV) and histamine.The abdominal aorta were processed and examined by HE stain.The levels of NF-Κb,MCP-1 and MMP1 were determined by means of immunohistochemistry,the protein expressions of NF-κB,MCP-1 and MMP1 in the lesions were examined by means of Western-blot.Results:Compared with Group B4,the thickness of cap in medication groups(group B1,B2and B3) was significantly thicker and the intima-media thickness was significantly decreased(P〈0.05).The ratio of thickness of the plaque cap and intima-media was significantly different between Group B4 and treatment groups(P〈0.05).Immunohistochemistry showed that expressions of NF-κB,MCP-1 and MMP-1 were found in the atherosclertic plaques.Compared with group B4,the expressions of NF-κB,MCP-1 and MMP-1 showed by immunohistochemistry in the group B2 and B3 were much lower(P〈0.05),but there was no significant difference between group B2 and group B3.The protein expressions of NFκB,MCP-1 and MMP1were found in the atherosclerosis lesions.Compared with group B4,the protein expressions of NFκB,MCP-1 and MMP1 in group B2 and B3 were much lower in the atherosclerosis lesions(P〈0.05).Compared with group B1,the expressions of NFκB,MCP-1 and MMP1 in the group B2 were lower(P〈0.05).Conclusion:NXTG has effects of anti-inflammation leading to stabilization of vulnerable plaques.
