摘要
目的:观察额尔敦-乌日勒(珍宝丸)对实验性家兔动脉粥样硬化(atherosclerosis,AS)形成的影响,并探讨其作用机制。方法:采用喂养高脂饲料构建家兔AS模型,将60只家兔随机分为6组:正常组,模型组,额尔敦-乌日勒高、中、低剂量组分别给予额尔敦-乌日勒0.4、0.2、0.1g.kg-1.d-1,阳性对照组给予辛伐他汀5mg.kg-1.d-1,连续喂养90天。在喂饲高脂饲料的同时灌胃给药,并于给药前1 d及开始后第30、60、90天分别取各组家兔空腹血检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)。90天后心脏采血,进行血液流变学检测,随后处死家兔,取主动脉做病理形态学检查。结果:与模型组相比,额尔敦-乌日勒中、高剂量组的血清TC、TG、LDL-C水平降低(P<0.05),额尔敦-乌日勒中、高剂量组主动脉斑块面积占总面积的百分比显著低于模型组(P<0.05);光镜显示额尔敦-乌日勒中、高剂量组主动脉内膜结构病变较模型组减轻,内膜/中膜厚度比小于模型组(P<0.05);血液流变学分析显示,额尔敦-乌日勒中、高剂量组全血黏度明显低于模型组(P<0.05)。结论额尔敦-乌日勒能抑制AS斑块的形成和发展,其作用机制可能与调节血脂、改善血液流变学有关。
Objective:To investigate the effects of eerdun- wurile on experimental atherosclerosis in rabbits., and study the meehanlsms. Methods: An atheroselerotic rabbit model was established by feeding high fat diet , sixty male rab- bits were randomly divided into six groups : control group, model group ( high fat diet) , high - dose Eerdun - wurilc trea- ted group(0.4kg -1 , d -t ) ,mid - dose - Eerdun - wurile treated group (0.2kg -1 , d -1 ) ,low - dose Eerdtm - wurile - treated group(0, lkg-l·d-l ) ,and simvastatin group(5mg·kg-1 · d-1. ), All Rabbits were fed for 90 days eonseeu- lively. Blood samples were collected at the 30~ days ,60th days , 90th days for examination of TC ,TG、LDL,HDL. At the 90th days, collect blood from heart,Test the blood flow velocity. Then Kin rabbits, Take the aorta and observe Pathologl- cal morphology. Results :High- dose- Eerdun- wufilo treated group and mid -dose - Eerdun- wufile treated group rabbits showed lower serum lipid levels than model group ( P 〈 0.05 ), high, mid - dose group were significantly lower ar- ea of atherosderotie to compare with model group(P 〈0, 05), Light migmseope examination found that. high,told- dose group intlmal thickness less than the model group ( P 〈 0.05 ), Heranrheology showed that the whole blood viscosity ( high shear, middle shear, low shear) lower than the model group( P 〈 O, 05 ). Concision : Eerdun - wurile could inhibit atherosclerotic formation and development The mechanism may be related to decreasing the blood lipid metsbolism and adiustinng blood flow velocity.
