利用木糖高产谷胱甘肽酵母菌株的诱变选育及培养条件优化 被引量:2

摘要 以CT1463(热带假丝酵母)为出发菌株,经紫外诱变结合镉盐平板筛选,获得一株高产谷胱甘肽的突变株CV26,其谷胱甘肽产量和含量分别为45.4 mg/L和7.05 mg/g,较出发菌株提高了41%和52%。对CV26发酵条件进行了优化,在优化发酵条件下谷胱甘肽产量和含量分别为128.70 mg/L和14.31 mg/g,分别为初始条件下的2.4倍和1.4倍。利用突变株CV26以玉米芯水解液为碳源进行GSH发酵实验,GSH产量和含量与纯木糖和模拟水解液发酵结果相近。 A mutant strain CV26 was selected by UV mutagenesis combined with cadmium sulfate resistance plate.The GSH yield and GSH content of CV26 were 45.4 mg/L and 7.05 mg/g,which ncreased by 41% and 52% respectively compared with original strain Candida tropicalis CT1463.Then the fermentation conditions were studied.Under the optimal fermentation condition,GSH yield and GSH content of mutant CV26 was 128.70 mg/L and 14.31 mg/g,showed 2.4 fold and 1.4 fold improvements over that of the initial condition.When with corncob hydrolysates as carbon source,the GSH yield and content of CV26 is equal to that under pure xylose condition.
