摘要
目的通过观察穴位埋线对单纯性肥胖大鼠摄食和脂代谢影响,探讨其减肥及降脂意义。方法高脂高糖饮食喂养10周制备单纯肥胖大鼠模型,随机分为正常组、模型组和埋线组。选取"三里""天枢"等穴埋置可吸收线,1次/7d,干预15d。检测大鼠体质量、日摄食量、血清总胆固醇(Total Cholesterol,TC)和低密度脂蛋白胆固醇(Low-Density Lipoprotein Cholesterol,LDL-C)水平、肝脏脂蛋白脂酶(Lipoprotein Lipase,LPL)和肝脂酶(Hepatic Lipase,HL)活性。用放射免疫分析法检测血清肥胖抑制素(Obestatin)含量,免疫组织化学法检测下丘脑肥胖抑制素阳性表达情况。结果与正常组比较,模型组血清TC和LDL-C水平显著升高(P<0.01),肝脏LPL和HL活性下降(P<0.05,P<0.01)。埋线组大鼠经埋线干预后体质量增长为负,较模型组大鼠体质量下降明显(P<0.01),日摄食量明显减少且摄食稳定,血清TC和LDL-C水平均下降(P<0.01,P<0.05),肝脏LPL和HL活性升高(P<0.05,P<0.01),血清肥胖抑制素含量高于正常组(P<0.01),但模型组和埋线组差异无统计学意义,模型组和埋线组大鼠下丘脑肥胖抑制素表达均高于正常组(P<0.01),且埋线组表达明显高于模型组(P<0.01)。结论穴位埋线能增强下丘脑和血清肥胖抑制素含量,从而抑制摄食;并且增强肝脏LPL和HL活性,加强血脂代谢。提示埋线可通过中枢调控机制,达到减肥目的,从而改善脂质代谢。
Objective To study the significance of catgut embedding at acupoints in reducing body weight and lipid by observing its effect on ingestion and lipid metabolism in rats with simple obesity.Methods A simple obesity model of rats was established by feeding rats with high fat and glucose diet for 10 weeks.Twenty-four SD rats with obesity were randomly divided into control group,model group and catgut embedding(CE) group,12 rats in each group.Rats in CE group were treated with embedded absorbable catgut at bilateral acupoints of Housanli,Tianshu,and Pishu,once a week.Rats in the other two groups were not interfered.Fifteen days after treatment,body weight and daily ingestion,serum levels of total cholesterol(TC) and low-density lipoprotein cholesterol(LDL-C),activities of lipoprotein lipase(LPL) and hepatic lipase(HL) were measured,respectively.Serum obestatin was assayed by radioimmunoassay and positive expression of obestatin in hypothalamus of rats was detected with immunohistochemical method.Results The serum TC and LDL-C levels were significantly higher while the activities of LPL and HL were significantly lower in model group than in control group(P〈0.01).The body weight was decreased greater,the daily food ingestion was less,the serum TC and LDL-C levels were significantly lower,and the activities of LPL and HL were significantly higher in CE group than in model group(P〈0.01).However,the serum obestatin level was higher in model group than in control group(P〈0.05) but with no difference between model and CE groups.The expression level of obestatin in hypothalamus of rats was higher in model and CE groups than in control group.However,the expression level of obestatin in hypothalamus of rats was significantly higher in CE group than in model group(P〈0.01).Conclusion Catgut embedding at acupoints can reduce body weight and lipid in rats with simple obesity by increasing the expression of obestatin in hypothalamus and serum of rats,thus inhibiting food ingestion,strengthening activities of LPL and HL in liver to accelerate serum lipid metabolism and decreasing serum TC and LDL-C levels.
