摘要
目的探讨整合素α6单克隆抗体(GoH3)对人乳头瘤病毒(HPV)6/11病毒颗粒(VP)体外感染HaCaT细胞的影响。方法①采用4组不同浓度的HPV6/11VP体外感染HaCaT细胞;②加入6组不同浓度的GoH3后,以10^6拷贝/mlHPV6/11vP体外感染HaCaT细胞;③加入稀释倍数为1:100的GoH3前后,应用8例临床分离的浓度为10e拷贝/ml的HPV6/11vP体外感染HaCaT细胞;④以上3步分别以荧光定量聚合酶链反应(FQ—PCR)方法检测感染后HaCaT细胞的HPVDNA载量并计算GoH3抑制率。结果①4组不同浓度的HPV6/11vP感染细胞后所测得的病毒载量不同(P〈0.01)。当HPV6/11VP浓度为10^6拷贝/ml时,HaCaT细胞被感染率为100%,且所测得的HPV载量高于其他3组(P〈0.05);②当GoH3稀释倍数〉1:100时,其对HPV6/11vP感染HaCaT细胞的抑制率与其浓度成正比;但当稀释倍数〈1:100时,再增加GoH3的浓度,其抑制率也未见明显增长;③8例临床分离的HPV6/11VP在加GoH3之前感染HaCaT细胞的平均病毒载量为(5.814-2.51)×104拷贝/ml,加入GoH3后感染HaCaT细胞的平均病毒载量为(3.02±1.21)×10^4拷贝/ml,计算GoH3的平均抑制率为(46.9±4.7)%。结论GoH3对HPV6/11 VP黏附到HaCaT细胞具有一定的抑制作用,推测整合素α6可能是HPV6/11VP感染宿主细胞的病毒受体之一。
Objective To investigate the influence of an anti-integrin α6 monoclonal antibody (GoH3) on the in vitro infection of a human keratinocyte cell line HaCaT with HPV6/11 virus particles (VP). Methods HaCaT cells were infected in vitro with 4 different concentrations of HPV6/11 VP alone, HPV6/11 VP of 106 copies/ml after incubation with 6 different dilutions of GoH3, or 8 clinical isolates of HPV6/11 VP of 106 copies/ml before or after the incubation with 1 : 100 dilution of GoH3. After additional culture, the infected HaCaT cells were collected and fluorescence quantitative (FQ)-PCR was performed to detect the HPV DNA load in cells. The inhibition rate of goH3 on the infection was calculated. Results The viral load was different among the HaCaT cells infected with different concentrations of HPV6/11 VP (P 〈 0.01). The inhibition rate on the infection positively correlated with the concentration of GoH3 when the dilution was more than 1 : 100; however, when the dilution was less than 1 : 100, the increase in GoH3 concentration had no influence on the inhibition rate. The average viral load in HaCaT cells infected with clinical isolates of HPV6/11 VP was (5.81 ± 2.51) ×10^4 copies/ml in the absence of GoH3, (3.02 ± 1.21) × 10^4 copies/ml with the presence of GoH3, and the average inhibition rate of GoH3 was (46.9 ±4.7)%. Conclusions GoH3 could partially suppress the adhesion of HPV6/11 VP to HaCaT cells, hinting that integrin oc6 is an important HPV6/11 VP receptor on host cells.
