摘要
目的探讨自噬相关基因Beclin1在喉鳞状细胞癌(简称喉癌)中的表达及其与临床病理特征的关系。方法采用免疫组织化学方法检测Beclin1蛋白在50例喉癌组织、45例癌旁组织、10例喉乳头状瘤及16例声带息肉组织中的表达情况,RT-PCR方法检测41喉癌组织、41癌旁组织和11例声带息肉组织中Beclin1 mRNA的表达情况。结果 Beclin1蛋白在喉癌组织、癌旁组织、喉乳头状瘤及声带息肉中的阳性率分别为58.0%、95.6%、90.0%和100%,Beclin1蛋白在喉癌组织中的表达率明显低于癌旁及声带息肉组织,具有显著的统计学差异(χ2=28.878,P<0.001)。Beclin1 mRNA在喉癌组织中的相对表达量为(0.47±0.022),显著低于癌旁组织(0.81±0.090)及声带息肉组织(0.80±0.041,F=334.968,P<0.05)。Beclin1蛋白在喉癌组织中的表达与肿瘤部位、组织学分级有关(P<0.05),与患者的年龄、T分级、临床分期及淋巴结转移无关(P>0.05)。结论 Beclin1基因在喉癌组织中表达下调,提示自噬活性的改变可能对喉癌的发生、发展起一定作用。
OBJECTIVE To Study the expression of autophagy gene Beclin1 in laryngeal squamous cell carcinoma(LSCC)and its clinical and pathological significance.METHODS The immunohistochemistry MaxVision method was used to detect expression of Beclin1 in 50 specimen of laryngeal carcinoma tissue,45 specimen of tissue adjacent to carcinoma,10 specimen of laryngeal papilloma tissue and 16 specimen of vocal cord polyp tissue.Reverse transcription-polymerase chain reaction(RT-PCR)method were used to detected the Beclin1 mRNA in 41 specimen of LSCC,41 specimen of tissue adjacent to carcinoma and 11 specimen of vocal cord polyp tissue.RESULTS The positive rate of Beclin1 was significantly lower in LSCC tissues than that in tissue adjacent to carcinoma and vocal cord polyp tissue(58.0% vs 95.6% and 100%,χ2=28.878,P0.001).The mRNA level of Beclin1 was significantly lower in LSCC tissues than in tissues adjacent to carcinoma and vocal cord polyp tissue(0.47±0.022 vs 0.81±0.090 and 0.080±0.01,F =334.968,P 0.05).The expression of Beclin1 had correlation with tumor location,pathological grade,but no relation to age,T stage,clinical stage and lymph node metastasis(P 0.05).CONCLUSION The expression of Beclin1 is down-regulated in laryngeal cancer tissue.The Beclin1 may play an essential role in carcinogenesis and development of laryngeal carcinoma.
