左旋甲羟戊酸对辛伐他汀调控心肌细胞PTEN表达的影响 被引量:1

摘要 目的:观察左旋甲羟戊酸(L-MVA)对辛伐他汀调控大鼠心肌细胞第十号染色体缺失的张力蛋白同源区(PTEN)表达的影响。方法:应用图像分析及[3H]-亮氨酸掺入法测定细胞表面积及蛋白合成速率,RT-PCR检测心钠素(ANP)mRNA表达,RT-PCR和Western blot法检测PTEN的mRNA和蛋白表达水平。结果:辛伐他汀+L-MVA组心肌细胞表面积、[3H]-亮氨酸掺入率、ANP mRNA水平均明显高于辛伐他汀组(P<0.01),而PTEN的mRNA及蛋白表达水平则明显降低(P<0.01)。结论:L-MVA能够降低PTEN的表达水平,逆转辛伐他汀对心肌细胞肥大的抑制作用。 Objective:To study the modulation of L-mevalonate(L-MVA) on the effects of simvastatin on PTEN expression in cultured rat cardiac myocytes.Methods: Image analysis system and -leucine incorporation were used to measure cell surface area and protein synthesis.The mRNA expression of atrial natriuretic peptide(ANP)was determined with RT-PCR.RT-PCR and Western blot were applied to study the mRNA and protein expression of PTEN,respectively.Results: Cell surface area,-leucine incorporation rate,ANP mRNA levels in simvastatin+L-MVA group were significantly higher than those of simvastatin group(P0.01),while the mRNA and protein levels of PTEN in simvastatin+L-MVA group were significantly lower than those of simvastatin group(P0.01).Conclusion:L-MVA can decrease the expression level of PTEN,thus reverse the inhibition effects of simvastatin on cardiac myocytes hypertrophy.
