摘要
以玉米自交系鲁9801、K10芽尖为受体,采用农杆菌介导法转化BcWRKY1转录因子基因,同时优化了遗传转化体系。结果表明,乙酰丁香酮浓度为100μmol/L、0.05个大气压下真空处理侵染10 min转化率最高。实验共筛选出98株除草剂抗性植株,其中23株PCR呈阳性,18株结实。
BcWRKY1 gene was introduced into 7 maize inbred lines by pollen tube pathway and optimize the influence factors of genetic transformation. The results indicated that the transformation method with 100 μmol/L AS in the medium combined with vacuum at 0.05 Mpa for 10 min was beneficial to improve the transformation rate. Ninety eight maize plants resisted to herbicide, among which 23 plants showed positive PCR reaction. Eighteen transformed plants were seed.
