摘要 目的观察TGF-β1对原代培养猪甲状腺细胞TPO表达的影响,探讨其可能的作用机制。方法体外培养猪甲状腺细胞,用MTT法测定TGF-β1对细胞存活率的影响。用愈创木酚法测定TGF-β1对细胞TPO活性的影响。实验分为0μg/L(对照组),2μg/L、5μg/L、10μg/L组。用RT-PCR方法检测甲状腺细胞TPO的mRNA表达,分析各处理组TPO基因表达的变化。结果①TGF-β1对猪原代甲状腺细胞存活率的影响:与对照组相比,2μg/L、5μg/L、10μg/L组细胞的存活率均明显降低(P<0.05)。②TGF-β1对TPO活性的影响:与对照组比较,2μg/L、5μg/L、10μg/L组染TGF-β1后,TPO活性均明显下降(P<0.05),呈显著的负相关。③RT-PCR结果:与对照组相比,给药各组的表达明显降低(P<0.05),并呈剂量-效应关系。结论 TGF-β1对猪原代培养甲状腺细胞有抑制的作用,并呈剂量-效应关系,其机制可能是与TGF-β1TPO基因的表达有关。 【Objective】To observe the effect of TGF-β1 on the expression of TPO mRNA in primary porcine thyrocytes,and approach its mechanism.【Methods】TGF-β1 injured model was made with purified cultured porcine thyrocytes,and the cell survival ratio was measured with MTT method and the TPO activity was measured with upgrade guaiacol method.the experiment was divided according to the final concentration of TGF-β1 into 0(control group),2,5,10μg/L.The TPO mRNA expression levels were determined by RT-PCR.Analyze the changes of gene expression among each treatment group.【Results】①Cell survival ratio,significant different between the groupes,with the increasing does of TGF-β1,level of cell survival ratio obviously lowered,having statiscal significance(P0.05);②With the dose of TGF-β1 increasing,TPO activity has a obviously lower and statistical significance in group comparison(P0.05).The TPO activity had a negative correlation with the dose of TGF-β1(P0.05).③With the dose of TGF-β1 increasing,the expression of TPO mRNA has a obviously lower and statistical significance in group comparison(P0.05).【Conclusion】It is an effect for thyroid function that TGF-β1 can reduced the expression of TPO mRNA and TPO activity so that level of thyroid hormone is decreased.
