有机碘和无机碘对人甲状腺细胞分泌功能影响的体外实验研究 被引量:2

Experimental Research on Effect of DIT and KI on Human Thyrocytes' Secretion in vitro

摘要 目的研究有机碘(DIT)和无机碘(KI)对体外培养的人甲状腺细胞(TEC)分泌三碘甲腺原氨酸(T3)、甲状腺素(T4)及甲状腺球蛋白(Tg)功能的影响。方法取正常甲状腺细胞进行原代细胞培养,将细胞密度调整为2×105/ml,加入DIT和KI溶液(含碘最终浓度分别为0、10-8、10-7、10-6、10-5、10-4、10-3 mol/L),培养48 h。采用放射免疫法测定细胞培养上清液中T3、T4及Tg的含量。结果与对照组比较,各剂量DIT组和KI组甲状腺细胞分泌的T3、T4和Tg含量均较高,差异有统计学意义(P<0.01)。当KI和DIT浓度均为10-5 mol/L时,T3含量升至最高;当KI浓度为10-5 mol/L或DIT浓度为10-4 mol/L时,T4含量升至最高;当KI和DIT浓度均为10-6 mol/L时,Tg含量升至最高。与相同剂量KI组比较,DIT浓度为10-6 mol/L时的T3含量较低,DIT浓度为10-5和10-4 mol/L时T4含量较低,DIT浓度为10-6和10-3 mol/L时的Tg含量均较低,差异有统计学意义(P<0.05或P<0.01)。结论适量碘可促进甲状腺细胞分泌T3和T4及Tg,DIT对甲状腺激素分泌的影响程度稍低于KI。 Objective To research the effect that organic iodine(DIT) and organism iodine(KI) on secretion of triiodothyronine(T3),thyroxine(T4) and thyroglobulin(Tg) of human thyroid cells in cultured.Methods The normal human thyroid epithelium cells which density were adjusted to 2×105/ml were cultured with different dose(0,10-8,10-7,10-6,10-5,10-4 and 10-3 mol/L) of DIT or KI for 48 hours.Radioimmunoassay was employed to determine the contents of T3,T4 and Tg in supernatant of thyrocytes in cultured.Results The contents of T3,T4 and Tg in all experimental groups were obviously higher than those in the control group(P0.01).When the doses of DIT and KI were both 10-5 mol/L,the content of T3 increased to the maximum.When the dose of DIT and KI were both 10-6 mol/L,the content of Tg increased to the maximum.Compared with those in KI groups with the same dose,the content of T3 in DIT group was lower at the dose of 10-6 mol/L DIT;the contents of T4 in DIT groups were lower at the doses of 10-5 and 10-4 mol/L DIT;The contents of Tg in DIT groups were lower at the dose of 10-6 mol/L and 10-3 mol/L DIT.Conclusion Moderate dose of iodine can contribute to thyrocytes' secretion of T3,T4 and Tg.The effect of DIT on secretion of thyroid hormone is less than that of KI.
