摘要
目的研究针刺神门和命门穴后大鼠中缝背核5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)含量变化,探讨针刺治疗失眠症的作用机制。方法将SD雄性大鼠18只随机分为对照组、针刺神门组、针刺命门组各6只,采用立体定位法将微透析探针置入大鼠中缝背核,运用微透析法收集大鼠针刺前40min,针刺后20、40、60、80、100、120min中缝背核细胞外液,用高效液相-电化学法(HPLC-ED)检测其5-HT和5-HIAA含量的变化。结果与针刺前相比,针刺大鼠神门和命门穴20min后中缝背核5-HT的水平显著下降(P<0.01),在其后2h维持在低水平;与对照组相比,大鼠针刺后2h各时相点中缝背核5-HT的水平也显著下降(P<0.01)。与针刺前的基础值及与对照组相比,针刺神门和命门穴均显著降低中缝背核5-HIAA含量(P<0.01)。结论针刺神门和命门穴能通过调节中缝背核组织中5-HT和5-HIAA含量从而起到调节睡眠的作用。
Objective To study the changes in the contents of 5hydroxytryptamine(5-HT) and 5-hydroxyindoleacetic acid(5-HIAA) of the dorsal raphe nucleus(DRN) of rats after acupuncture at Shenmen(HT 7) and Mingmen(GV 4) to discuss the mechanism of acupuncture treatment for insomnia.Methods Eighteen SD male rats were randomized into control group,acupuncture Shenmen(HT 7) group and acupuncture Mingmen(GV 4) group,with 6 in each.A microdialyzer probe was placed into the DRN with stereotaxic technique.The extracellular fluid of DRN 40min before acupuncture and 20,40,60,80,100,and 120min after acupuncture was collected by microdialysis,and the changes in 5-HT and 5-HIAA contents were determined with high performance liquid chromatography-electrochemical detection(HPLC-ED).Results Compared with before acupuncture,the content of 5-HT of DRN was significantly decreased 20min after acupuncture at Shenmen(HT 7) and Mingmen(GV 4)(P〈0.01),and thereafter it remained low within 2 hours.Compared with the control group,the content of 5-HT of acupuncture groups 2h after acupuncture at each time point was also significantly decreased(P〈0.01).Compared with the basic value before acupuncture and with the control group,acupuncture at Shenmen(HT 7) and Mingmen(GV 4) both could decrease the content of 5-HIAA of DRN(P〈0.05).Conclusion Acupuncture at Shenmen(HT 7) and Mingmen(GV 4) functions to regulate sleep through regulating the contents of 5-HT and 5-HIAA in the tissues of DRN.
