摘要
目的:观察散瘀止痛方熏洗对于角叉菜致炎后大鼠足趾肿胀度的影响及对致炎足局部组织前列腺素E2(PGE2)含量及环氧合酶-2(COX-2)表达的影响。方法:24只SD雄性大鼠,用角叉菜造模后随机分为四组:生理盐水组、散瘀止痛方Ⅰ组、散瘀止痛方Ⅱ组、塞来昔布组。将造模后4组大鼠,同时予灌胃及浸泡。观察致炎后30m in,1h,2h,3h的足趾肿胀度。致炎3h后,处死大鼠,采集标本。用ELISA法测定足趾局部组织的PGE2含量,用RT-PCR法测定足趾局部组织的COX-2 mRNA的表达。结果:与生理盐水相比,散瘀止痛方能显著抑制大鼠足趾肿胀度,能降低前列腺素E2(PGE2)含量(P<0.01)及抑制环氧合酶-2(COX-2)mRNA的表达(P<0.01)。结论:散瘀止痛方熏洗抗炎止痛机理可能与降低前列腺素E2含量及抑制COX-2的表达有关。
Objective: To observe the effect of the carrageenan-induced rat paw swelling and the contents of PGE2 and the expression of COX-2 by Fumigation and Washing with Sanyuzhitong Decoction.Methods: After building the animal model of "inflammatory edema and pain",which was induced by carragheen,24 SD rats were randomly divided into 4 group(each group had 6 rats),which were normal saline group,Sanyuzhitong DecoctionⅠgroup Sanyuzhitong DecoctionⅡgroup and Celecoxib group.Rats in each group were administered by soaking and gavage.All the rats were soaked for 30 minute.Then the thickness of the inflamed paw was measured respectively at 30 minute,1hour,2hour and 3hour,and the degree of swelling was calculated.After that,the rats were killed and the specimens were collected.The contents of PGE2 were determined by ELISA.The expression of COX-2 was determined by RT-PCR.Results: The degree of swelling in Sanyuzhitong DecoctionⅠ group,Sanyuzhitong DecoctionⅡ group and Celecoxib group were obviously lower than that in normal saline group.The contents of PGE2 and the expression of COX-2 in Sanyuzhitong DecoctionⅠ group,Sanyuzhitong DecoctionⅡ group and Celecoxib group were obviously lower than that in normal saline group.Conclusion:The mechanism of anti-inflammation and analgesic effect with Sanyuzhitong Decoction may be by reducing the local contents of PGE2 and inhibiting the expression of COX-2.
