摘要
目的:探讨白桦酯醇对人卵巢癌裸鼠移植瘤凋亡作用的影响。方法:建立人卵巢癌裸鼠移植瘤模型,将16只裸小鼠随机分为对照组、白桦酯醇组,观察并记录各组裸鼠及其移植瘤的生长情况,计算肿瘤体积。免疫组化检测各组裸鼠皮下移植瘤中P53、Bax、Bcl-2及凋亡指数的表达水平。透射电镜下观察肿瘤组织的超微结构。结果:①白桦酯醇组裸鼠移植瘤的体积较对照组明显变小(P<0.01)。②白桦酯醇组的P53、Bax表达较对照组均明显增高(P<0.01),与凋亡指数变化呈同向变化趋势,Bcl-2表达较对照组无明显变化(P>0.05)。③白桦酯醇可诱导裸鼠移植瘤细胞的凋亡。结论:白桦酯醇对裸鼠移植瘤有明显抑制作用,可能与肿瘤细胞的凋亡作用有关。
Objective:To investigate the inhibitory and apoptotic effect of betulin on human ovarian cancer nude mice transplanted tumor.Methods:Human ovarian cancer nude mice transplanted tumor models were established,and 16 nude mice were divided into 2 groups:the control group and the betulin group.The changes of tumor volumes were recorded.Also the expression of P53、Bax、Bcl-2 and the apoptotic index were determined by immunocytochemistry(IC).The ultramicrostructures of the tumor tissues were observed by transmission electron microscope.Results:①Tumor volumes in the experimental group were much smaller than those in the control group(P0.01);②The expressions of P53、Bax in the experimental group were significantly higher than those in the control group(P0.01),also the same tendency was found in apoptosis between the two groups.The expression of Bcl-2 in the experimental group was not markedly changed compared with that in the control group(P0.05);③Betulin could induce the apoptosis of the tumor cells in nude mice concentration dependently.Conclusions:Betulin has obvious inhibition function on the growth of ovarian cancer tumor,which might be mediated by apoptosis.
