摘要
目的 用重组质粒pc D N A3 - P30 免疫 B A L Bc 小鼠, 观察其 D N A 免疫所诱导的小鼠体液免疫反应和保护性作用。方法 大量制备质粒 D N A, 肌肉注射免疫小鼠。 E L I S A 测定 Ig G 抗体滴度; 免疫鼠血液组织 P30 基因 P C R 扩增; 弓形虫攻击感染。结果 用 E L I A S 法检测的抗体 Ig G 滴度1∶2560 , 免疫后3 周及6 个月从免疫鼠血液组织中检测到 P30 基因;攻击感染实验组的存活时间较对照时间延长( P< 005) , 结论 重组质粒pc D N A3 - P30 免 B A L Bc 小鼠可诱导一定的体液免疫应答, 对抗攻击感染具有一定的保护性。
AimDirectly immunizing BALB/c mice with the recombinant plasmid with the P30 gene of Toxoplasma gondii,to observe the humoral and protective immune responses induced by pcDNA3-P30.MethodsLarge scale preparation of the recombinant plasmid pcDNA3-P30,it was injected into the BALB/c mice by muscles.The IgG antibodies in serum from vaccinated mice were detected by anzyme-linked immunosorbent asssy(ELISA).The P30 gene was detected in the blood of immunized mice by PCR method at 6th month after last injection.The vaccinated mice were challenged by RH tachyzoit of Toxoplasma gondii.ResultsThe titer of specific IgG antibody in serum from vaccinated mice have been shown at 1∶2560.The P30 gene was detected in the blood of immunized mice by PCR at 3 rd and 6th month after last injection.The survival time of immunized mice was longer than that of control group(P<0 05)after being challenged with.Toxoplasma gondii.ConclusionDNA vaccine with the recombinant plasmid pcDNA 3-P30 could elicit BALB/c mice to generate numoral and protective immune responses for Toxoplasma gondii infection. \ \
