N-乙酰半胱氨酸对H_2O_2刺激的支气管上皮细胞表达IL-8影响

摘要 目的:探讨N-乙酰半胱氨酸(NAC)对过氧化氢(H2O2)诱导的支气管上皮细胞分泌白介素-8(IL-8)影响。方法:将6例人支气管上皮细胞培养传代后分为:对照组,H2O2组,NAC+H2O2组。NAC+H2O2组中先给予NAC预孵育4 h之后给予H2O2。通过MTT法检测NAC和H2O2对细胞作用合适浓度,通过ELISA法检测人支气管上皮细胞上清中IL-8的含量,并检测细胞中谷胱苷肽(GSH)的含量,通过凝胶迁移率阻滞实验(EMSA)检测核转录因子-κB(NF-κB)、激活蛋白-1(AP-1)的表达。结果:(1)H2O2组中GSH含量较对照组明显减少,NAC+H2O2组GSH较H2O2组明显升高(P<0.01)。(2)H2O2组中IL-8含量较对照组明显增加,NAC+H2O2组较H2O2组明显降低(P<0.01)。(3)H2O2组中转录因子NF-κB、AP-1的活性明显增强,NAC+H2O2组NF-κB、AP-1的活性较H2O2组明显下调。结论:NAC可增强支气管上皮细胞的抗氧化能力,并通过下调NF-κB、AP-1活性而抑制IL-8的合成。
