不同浓度苦参碱对神经母细胞瘤SH-SY5Y细胞存活率及凋亡率的影响 被引量:5

摘要 目的探讨不同浓度苦参碱对神经母细胞瘤SH-SY5Y细胞存活率及凋亡率的影响。方法分别应用MTT法和FCM法检测苦参碱与顺铂对NB细胞株SH-SY5Y存活率的影响。结果 MTT法检测SH-SY5Y细胞的存活率(%)分别为:(90.75±6.94)、(88.61±10.22)、(55.31±1.83)、(77.37±2.24)、(61.21±7.92)、(44.71±9.51)。FCM法检测SH-SY5Y细胞的凋亡率(%)分别为:(6.89±1.03)、(7.63±1.38)、(16.36±2.57)、(8.42±1.76)、(14.79±2.37)、(18.46±2.44)。除经ATRA诱导的顺铂干预组外,各组存活率及凋亡率与对照组比较差异均有统计学意义(P<0.05);未经ATRA诱导的顺铂干预组存活率及凋亡率与经ATRA诱导的顺铂干预组比较差异有统计学意义(P<0.05);苦参碱0.5mg/mL组与经ATRA诱导的顺铂干预组比较,差异无统计学意义(P>0.05)。结论苦参碱可以抑制神经母细胞瘤SH-SY5Y细胞系增殖、诱导凋亡,可以提高顺铂化疗的敏感性,在本实验范围内浓度越高这种抑制增殖、诱导凋亡作用越明显。 Objective To explore the effects of different concentration of Matrine on NB in survival and apoptosis of SH-SY5Y cells.Methods MTT assay and FCM assay were performed to exzamination the changing of cell survival rate of SH-SY5Y cells by the combined action different concentration of Ma and constant concentration of CDDP separately.Results The MTT method detection the cell of SH-SY5Y survival rate below:90.75±6.94;88.61±10.22;55.31±1.83,77.37±2.24,61.21±7.92,44.71±9.51;FCM method to detect apoptosis rate below:6.89±1.03,7.63±1.38,16.36±2.57,8.42±1.76,14.79±2.37,18.46±2.44.There was significant difference in survival and apoptosis of SH-SY5Y cells between each group and matched group except the group of cisplatin which induced by ATRA;and also between uninduced group of cisplatin and the group of cisplatin which induced by ATRA (P 0.05);The survival rates and apoptosis of SH-SY5Y cells was statistically significant between the group of Matrine and the group of Matrine joint cisplation(P 0.05);and also between the group of Matrine joint cisplation and uninduced group of cisplation(P 0.05);There was no significant difference in survival and apoptosis of SH-SY5Y cells between Matrine(0.5mg/mL)and the group of cisplatin which induced by ATRA (P 0.05).Conclusion Matrine can induce cell apoptosis and inhibit the growth of the SH-SY5Y cells,meanwhile increasing the sensibility of chemotherapy of cisplatin;in this lab,the action of cytostatic and the induced apoptosis was obviously with the higher concentration.
