摘要
目的建立番石榴叶中金丝桃苷的HPLC含量测定方法。方法采用Dikma Kromasil C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.1%磷酸溶液(45∶55)为流动相,流速为1.0 mL.min-1,检测波长为254 nm。结果金丝桃苷在0.020 48~0.204 8μg内呈良好的线性关系(r=0.999 9),平均回收率为98.4%,RSD为1.5%。结论本方法简单、准确、重复性好、专属性强,可用于番石榴叶药材及制剂中金丝桃苷的含量测定。
OBJECTIVE To establish a determination method for hyperin in Psidium guajava leaf by HPLC. METHODS HPLC was performed on a Dikma Kromasil C18 column(4.6 mm×250 mm, 5 μm) with methanol-0.1% phosphoric acid solution(45∶55) as the mobile phase at a flow rate of 1.0 mL.min-1, and the detection wavelength was 254 nm. RESULTS It showed a good linearity at the range of 0.020 48~0.204 8 μg(r=0.999 9), and the average recovery of hyperin was 98.4%(RSD= 1.5%). CONCLUSION The method is sensitive, accurate, reproducible, specific and could be used to determine hyperin in Psidium guajava leaf.
