摘要
目的制备超顺磁性Fe3O4纳米粒(superparamagnetic Fe3O4 nanoparticle,SPFN),探讨其对小鼠的急性毒性作用。方法采用化学共沉淀法制备SPFN,应用透射电镜和振动样品磁场计等对SPFN进行形态观察和结构表征测定。取60只小鼠按不同的给药方式和剂量,随机数字表法分为3组(n=20):口服灌胃组(总剂量:2 104.8 mg/kg、容积:25 ml/kg)、腹腔注射组(总剂量:1 578.6 mg/kg、容积:20 ml/kg)、尾静脉注射组(总剂量:438.5 mg/kg、容积:10 ml/kg);每组另取10只小鼠予生理盐水作为对照(n=10)。观察小鼠的一般情况;全自动生化分析仪检测小鼠主要血生化指标;HE染色观察主要脏器的病理改变;瑞氏染色观察骨髓组织。结果制成粒径为15~20 nm,饱和磁化强度为21.431 emu/g,剩余磁化强度为0.102 emu/g的SPFN。予SPFN 2周后,各组小鼠均未死亡,血生化指标均无统计学差异(P>0.05),主要脏器和骨髓均未见炎症、水肿、变性、坏死等改变。结论成功制备超顺磁性Fe3O4纳米粒,自制超顺磁性Fe3O4纳米粒对小鼠无明显急性毒性。
Objective To study the acute toxicity of superparamagnetic Fe3O4 nanoparticles(SPFN) to mice.Methods The morphology and structural characteristics of SPFN prepared by chemical co-precipitation were determined with a vibratory sample magnetometer under transmission electron microscope.Sixty mice were randomly divided into oral enema group(a total dose of 2 104.8 mg/kg and a volume of 25 ml/kg),intra-peritoneal injection group(a total dose of 1 578.6 mg/kg and a volume of 20 ml/kg),and caudal vein injection group(a total dose of 438.5 mg/kg and a volume of 10 ml/kg),20 mice in each group,according to different drug administration methods and dosages.Ten mice in each group that received the same dose of normal saline via the same way served as controls.General condition of the mice was observed during experiment,major blood biochemical indicators were analyzed with an automatic biochemical analyzer,and pathological changes in major organs and myeloid tissue were observed with HE staining and Wright staining,respectively.Results The diameter of prepared SPFN was 15-20 nm with a saturated magnetization of 21.431 emu/g and a remanent magnetization of 0.102 emu/g.Two weeks after administration of SPFN,no mice died,no significant difference was found in blood biochemical indicators among different groups,and no inflammation,edema,degeneration,necrosis were observed in major organs and bone marrow.Conclusion The SPFN we prepared have no acute toxicity to mice.
