摘要
目的研究不同剂量蜕皮甾酮(EDS)对人脐带间充质干细胞体外增殖的影响。方法分离培养并鉴定人脐带间充质干细胞(hUCMSCs)。取第3代hUCMSCs分别在基础培养基(低糖DMEM培养基、10%的胎牛血清、100U/m青/链霉素、4mmol/L L-谷氨酰胺)中加入不同浓度(0、25、50、100、150、200μg/ml)EDS予以干预,分别于第1、3、5、7天行MTT法检测细胞增殖活性;另外,设置相同分组EDS干预培养hUCMSCs7天,绘制细胞生长曲线。结果第3代细胞阳性表达CD29、CD105,阴性表达CD34、CD45。MTT法检测结果显示经EDS干预培养的hUCMSCs增殖能力较空白对照组有明显差异(P<0.05),其中EDS100、150及200μg/ml三组细胞活性比较无显著差异(P>0.05),较其他实验组有统计学意义(P<0.05)。细胞生长曲线同样显示EDS实验组可有效促进间充质干细胞的增殖。结论一定浓度范围内EDS对hUCMSCs具有促增殖作用,该作用在EDS浓度为100μg/ml时较为显著,不随浓度的增加而增强。这有望成为促进脐带间充质干细胞增殖的新方法。
Objective To investigate the effect of ecdysterone on the proliferation of human umbilical cord mesenchymal stem cells(hUCMSCs) in vitro.Methods hUCMSCs isolated by enzyme digestion from human umbilical cord tissues were cultured and identified for the surface antigens using fluorescence-activated cell sorting(FACS) .The cells were treated with ecdysterone at the concentrations of 0,25,50,100,150,and 200 μg/ml,and the changes in the cell proliferation were detected using MTT assay.Results The third-passage hUCMSCs were positive for CD29 and CD105 and negative for CD34 and CD45 as shown by flow cytometry.Treatment with ecdysterone resulted in significantly increased cell proliferation as compared to the control cells(P0.05) ,but no significant differences were found in cells treated with 100,150,and 200 μg/ml ecdysterone(P0.05) .The growth curves of the cells also demonstrated the definite effect of ecdysterone in promoting the proliferation of hUCMSCs.Conclusion Ecdysterone can promote the proliferation of hUCMSCs in vitro with the optimal concentration of 100 μg/ml,suggesting its potential value in the enrichment of mesenchymal stem cells.
