摘要
目的探讨醇溶性固定液在碱性磷酸酶(alkaline phosphatase,ALP)染色中的应用。方法将60例新鲜标本各取2块组织设为A、B两组,A组冷丙酮固定,B组醇溶性固定液固定,两组均常规石蜡切片,同时设立对照组,进行碱性磷酸酶钙-钴法染色,并对染色结果进行观察。结果两组切片均可见黑色碱性磷酸酶存在,但B组的阳性强度明显高于A组,对照组呈阴性反应。结论醇溶性固定液在对碱性磷酸酶的固定效果和显示方面,均优于冷丙酮固定液。
Objective To explore the application of alcohol soluble fixing solution in alkaline phosphatase staining.Methods Two tissues in each of 60 fresh samples were taken and assigned as group A and group B fixed by cold acetone and alcohol soluble fixing solution respectively.Conventional paraffin slice were obtained for the two groups and the corresponding control group was set.All groups underwent alkaline phosphatase Ca-Co staining.Results Black alkaline phosphatase were observed in groups A and B,the positive intensity in group B was higher,and the control group was negative.Conclusion Alcohol soluble fixing solution is better than cold acetone in fixing and staining alkaline phosphatase.
