摘要
①目的筛选出一种适合提高刺五加苷含量的内生镰刀霉菌P109-4生长的培养基。②方法将P109-4接种于6种基础培养基中,30℃下培养,对其生长状况进行观察,并在12、24、36、48、60、72h时段测量菌落大小。筛选出一种长速率最快的的培养基。再对其进行改良找到能使P109-4生长最快的培养基。③结果 P109-4在6种基本培养中生长状况明显不同,30℃培养72h后内生菌在牛肉膏蛋白胨、高氏一号、查氏培养基、PDA培养基、乳糖牛肉膏蛋白胨培养基、LC培养基的生长直径分别为4.00、3.70、3.75、3.90、3.20、3.20、3.63 cm。在改良牛肉膏蛋白胨培养基试验中,培养72h后牛肉膏蛋白胨培养基、改良牛肉膏蛋白胨培养基1号、改良牛肉膏蛋白胨培养基2号生长直径分别为3.30、3.45、3.20 cm。④结论经过72h菌落直径的比较,刺五加内生镰刀霉菌在改良牛肉膏蛋白胨培养基1号,生长速率更快。
Objective In order to select a culture medium that is more adapted to the growth of endogenous fungis P109-4.Methods Meeting the endogenous fungi to 6 kinds of based culture mediums and culture in 30℃.According to observe the develop situation and measure the diameter of the clones in 12h、24 h、36 h、48 h、60 h、72 h,select which culture medium of endogenous fungis P109-4 develop fastest.Results The develop situation of endogenous fungis P109-4 is different in 6 culture medium,After 72 hours the colony diameters are 4.00cm、3.70cm、3.75 cm、3.90 cm、3.20 cm、3.20 cm、3.63 cm in 6 culture mediums.In beef extract and peptone,culture medium improved assay.After 72 hours,the colony diameters are 3.30 cm、3.45 cm、3.20cm in 3 culture mediums.Conclusion Compare of the colony diameters after 72 hours,the NO.1 improved beef extract and peptone culture medium is most adapted to the development of endogenous fungis of Eleutherococcus senticosus.
