摘要
目的:应用蛋白质组学方法来比较早期肺鳞癌患者和健康人血清蛋白质的差异,筛选与肺鳞癌发生发展密切相关的蛋白质。方法:以6例Ⅰ期肺鳞癌患者血清和健康正常人血清为研究对象,运用比较蛋白质组学方法,用双向凝胶电泳(2-Pi-mentional electrophoresis,2-DE)分离肺鳞癌患者血清和正常人血清的总蛋白质,银染显色后经图像分析识别差异表达的蛋白质,再应用基质辅助激光解吸电离飞行时间质谱(Matrix-assisted laser desorption/ionization time offlightmass spectrometry,MALDI-TOF-MS)鉴定差异表达的蛋白质点。结果:肺鳞癌患者血清和健康正常人血清的平均蛋白质点数分别为939±40和962±27,匹配率达89.2%和87.1%;找到表达量差异3倍以上蛋白质点25个;对25个差异蛋白质点进行分析,得到了16张肽质量指纹图谱,最终鉴定出8个差异蛋白质。其中,有3个蛋白质只在早期肺鳞癌患者血清中表达,1个蛋白质在早期肺鳞癌患者血清中表达显著上调。另4个蛋白质在早期肺鳞癌患者血清中表达显著下调或缺失。结论:通过对早期肺鳞癌患者和健康正常人血清的蛋白质表达谱进行比较,鉴定出8个与早期肺鳞癌可能相关的蛋白质,为进一步筛选肺鳞癌早期诊断、治疗和预后评估的分子生物学标志物奠定了基础。
Objective:To compare and analyze the difference in serum proteins between patients with early-stage lung squamous carcinoma(LSC)and normal subjects and identify the proteins related with LSC.Methods:Serum samples were collected from 6 normal subjects and 6 patients with stage 1 of LSC.2-DE were performed to separate serum proteins.After silver staining,the proteins of differential expression were analyzed and then identified by matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS).Resluts:There were an average of(939±40)spots and(962±27)spots on 2-DE matching of two groups and the matching rates were 89.2 % and 87.1%.From the two groups,25 differential protein spots with over three times of differential expressions were analyzed by MALDI-TOF-MS.8 proteins were identified,of whith 3 proteins were expressed only in sera of the patients with early-stage LSC.Up-regulated expression was observed in 1 protein and down regulated expression or scarce expresseion were observed in 4 proteins in the patient serum group.Conclusion:Compared with the normal subject group,the sera in the early-stage lung squamous carcinoma patients group showed differential expression in protein profile.The proteins with differential expression are expected to be the serum markers for lung squamous carcinoma.
