摘要
本研究通过单因素试验及响应面分析试验优化了酸性蛋白酶水解鳗鱼的最优条件:加酶量19766 U.g-1,底物蛋白浓度15.28%,水解温度45℃,pH 3.0.在此条件下,可通过改变水解时间来控制水解度,从而制备不同工艺要求的水解产物.当水解时间为8 h时,水解反应达到平衡,水解度为33.35%,总氮回收率为80.63%.
The effects of the main parameters including hydrolysis temperature,pH value,hydrolysis time were investigated with single factor experiment and dosage of enzyme,protein concentration were investigated with response surface methodology(RSM).The optimum enzymatic hydrolysis conditions were as follows: the protein substrate with a final concentration of 15.28% was mixed with acid protease with a final concentration of 19766 U per gram of protein at pH 3.0.After incubating at 45 ℃ for 8 h,the degree of hydrolysis was 33.35%,and the recovery of total nitrogen was 80.63%.
