摘要
目的探讨有效体外构建高质量组织工程软骨的细胞种植技术。方法将分离、培养的兔第1代软骨细胞分别通过纤维蛋白凝胶种植技术(实验组)及常规种植方法(对照组)接种于聚乳酸-羟基乙酸共聚物(PLGA)支架。体外培养3周后收获组织工程软骨,行大体观察,组织切片,定量检测氨基糖胺聚糖(GAG)、DNA含量及Ⅱ型胶原染色面积。结果实验组较对照组构建的组织工程软骨体积更大、更有弹性和光泽;其Ⅱ型胶原染色面积、GAG、DNA含量均高于对照组(P<0.05)。结论与传统种植方法比较,采用纤维蛋白凝胶种植技术能在体外形成更高质量的组织工程软骨。
Objective To explore the effective technique to improve seeding efficiency in the construction of engineered cartilage in vitro.Methods Rabbit chondrocytes and poly(lactideco-glycolide)(PLGA) scaffold were employed to construct engineered cartilage tissue in vitro using fibrin gel(group A) and the conventional seeding technique(group C).After cultured for 3 weeks,the engineered cartilage was harvested and examined by gross observation,histological staining and immunohistochemisty of collagen Ⅱ.The contents of GAG,DNA and the percentage of collagen Ⅱ dyeing area were determined.Results Compared to group C,the engineered cartilage in group A had a bigger mass with more smooth and glossy surface and contents of GAG,DNA and the percentages of collagen Ⅱ(P〈0.05).Conclusion Compared to conventional method,the fibrin gel technique is an effective way for seeding cells onto scaffolds and getting high-quality engineered cartilage in vitro.
