七叶皂苷钠对大鼠局灶性脑缺血/再灌注损伤脑的保护作用 被引量:4

摘要 目的探讨七叶皂苷钠对大鼠局灶性脑缺血/再灌注损伤脑的保护作用。方法198只健康雄性Wistar大鼠,用大脑中动脉线栓法建立局灶性脑缺血/再灌注大鼠模型,随机分为假手术组、缺血/再灌注组和缺血/冉灌注七叶皂苷钠治疗组,假手术组18只,后两组又分为缺血2 h后再灌注6、12、24、48和72h,共5个时间点,每时间点18只。进行各组神经功能缺损评分,测定梗死体积,观察病理形态学变化,并用SP法和图像分析测定其缺血区c-fos基因的表达变化。结果缺血/冉灌注组c-fos基因在神经元的表达于再灌注6h开始增加,24h达高峰,48h后降低,72h最低,缺血/再灌注七叶皂苷钠治疗组c-fos基因于再灌注同一时相显著减少。结论七叶皂苷钠能减轻大鼠脑组织的缺血/再灌注损伤,改善神经功能缺失,其作用机制可能与减少c-fos基因表达有关。 Objective To study the effect of sodium aescinate on the c-fos expression in rats with cerebral ischemia reperfusion injury and to discuss its nerve protection mechanism.Methods One hundred and ninety-eight healthy male Wistar rats were chosen to establish focal cerebral ischemia reperfusion models with middle cerebral artery thread embolism method and were randomly divided into sham operation group,ischemia/reperfusion group,ischemia and reperfusion treated with sodium aescinate groups.There were eighteen rats in sham operation group.The other two groups were further divided into five subgroups according to reperfusion time interval of 6, 12,24,48,72 h after brain ischemia,with eighteen rats in each subgroup.To observe neurologic scores,infarct size,change of pathomorphology and the c-fos expression in brain tissue were measured with immunohistochemical method image analysis.Results The expression of c-fos started to increase at 6 h,reached the peak at 24 h,decreased at 48 h,and reached the lowest at 72 h in cerebral ischemia and reperfusion group.At the same time,the expression of c-fos in the cerebral ischemia and reperfusion treated with sodium aescinate group obviously decreased. Conclusion The c-fos expression may be one of the mechanism which induce neuronal apoptosis after cerebral ischemia and reperfusion,and sodium aescinate can help to recover the damaged brain by decreasing c-fos expression after early brain ischemia and reperfusion.
