RP-HPLC法测定五苓散中麦角甾醇和桂皮醛的含量

摘要 目的建立五苓散中麦角甾醇和桂皮醛的含量测定方法。方法五苓散样品经甲醇超声提取后,用HPLC法测定。色谱柱为HypersllC18(250mm×4.6mm,5μm),流动相为甲醇-水梯度洗脱,流速为1mL.min-1,检测波长为283nm,柱温为30℃。结果麦角甾醇在0.0437~1.3098mg.mL-1浓度范围内线性关系良好(r=0.9991),平均回收率为102.9%,RSD为1.5%(n=6);桂皮醛在0。0062~0.1872mg.mL-1浓度范围内线性关系良好(r=1.0000),平均回收率为97.8%,RSD为1.9%(n=6)。结论该方法简便、准确、可靠,可用于五苓散中麦角甾醇和桂皮醛的同时含量测定。 OBJECTIVE To establish a RP-HPLC method for quantitative determination of ergosterol and cinnamaldehyde in Wulingsan.METHODS Wulingsan was extracted with methanol by ultrasound extraction.The separation was performed on a Hypersil C18 column(4.6mm×250mm,5μm) at 30℃ using a gradient elution consisting of mobile phase A(methanol) and mobile phase B(water).The detection wavelength was 283nm.RESULTS Ergosterol showed a good linear relationship at the range of 0.0437~1.3098mg·mL-1,and cinnamaldehyde was from 0.0062 to 0.1872mg·mL-1.The average recovery of ergosterol was 102.9%(RSD=1.5%,n=6),cinnamaldehyde was 97.8%(RSD=1.9%,n=6).CONCLUSION The developed method is simple,accurate and reliable with good repeatability,and can be used for routine analysis of ergosterol and cinnamaldehyde in Wulingsan.
