The Relation between Improved Wound Healing in Diabetic Rats by Platelet-derived Wound Healing Factors and Increased Gene Expression of Procollagen Ⅰ (α_1)

摘要 目的 探索外源性血小板源伤口愈合因子 (platelet -derivedwoundhealingfactors,PDWHF)对糖尿病因难愈性伤口愈合的改善作用及其相关的分子机制。方法 四氧嘧啶诱导SD大鼠血糖值 >1.8mg/L后 ,采用大鼠背部成对切口伤模型 ,一侧伤口局部应用PDWHF( 10 0 μg/切口 ) ,手术当日及术后连续 6日每日 1次 ,另一侧作为自身对照。体外培养肉芽组织成纤维细胞 ,生长融合后 ,加入 10 %PDWHF ( 5g/L) ,观测前胶原Ⅰ (α1)mRNA水平量。结果 伤后 7,10 ,14天治疗组与自身对照非治疗组伤口抗张力值分别为 ( 10 3± 13 ) ,( 2 4 9± 4 9) ,( 5 4 6± 3 7)g/cm和 ( 67± 12 ) ,( 113± 2 8) ,( 2 77± 3 0 )g/cm ,两组间差异显著 (P <0 .0 1)。体外实验发现 :生长融合的伤口成纤维细胞与PDWHF孵育 4 ,8,12小时后 ,PDWHF作用组前胶原Ⅰ (α1)mRNA分别较对照组高 0 .9,3 .7,2 .2倍。结论 PDWHF促进糖尿病大鼠伤口愈合与其直接刺激伤口成纤维细胞前胶原Ⅰ (α1)基因表达相关。 Objective Impaired wound healing in diabetic subjects has been demonstrated to be associated with abnormality in the sort and number of multiple growth factors. This study was designed to determine PDWHF- (platelet-derived wound healing factors) induced acceleration of impaired healing of wounds in diabetic rats and the molecular mechanisms involved. Methods Two parallel incisions of 4cm were made on the dorsal of diabetic rats induced by Alloxan. PDWHF of 100μg (5 g/L) was applied to either of the incisions after operation, then once a day for next 6 days on run. The other incision was served as the control. 10% PDWHF (5 g/L) was added to the wound fibroblasts of confluence growth. The levels of mRNA of procollagen Ⅰ(α l) were detected at 4, 8 and 12 hours after addition of PDWHF.Results The wound breaking strength in the treated group amounted to 103±12 g/cm, 249±49 g/cm and 546±37 g/cm at post-wounding days 7, 10 and 14, respectively. In contrast, in the control it was only 67±12 g/cm, 113±28 g/cm and 277±30 g/cm, respectively. There was significant difference between the two groups ( P <0.01). The levels of mRNA of procollagen Ⅰ(α l) in PDWHF-treated wound fibroblasts were observed to be 0.9, 3.7 and 2.2 folds higher than those in the control at 4, 8 and 12 hours, respectively, afte raddition of PDWHF. Conclusions PDWHF could obviously improve the impaired healing of wounds in diabetic rats, which is related to the increased gene expression of procollagen Ⅰ(α l).
